How to make my pen drive read only?

Some flash drives such as the PNY attatche' Have a tiny physical switch that makes then read only- You flick it and you cant write to the drive.
I have only seen this function on the PNY attatche' flash drives though and there quite 'old' drives and may not be available in capacity's they you require.
There are 'newer' drives that some times come with security software on them but this is mainly to stop 'you' from seeing the data on the drive it dosent make them read only.
You can buy a hardware USB write blocker or the free way is to use a software write blocker that uses Windows XP's (with Service Pack 2) USB write blocking feature. All it does is change a value in a registry key. You can do this with group policy for your network.
